I also have a 10 1/2” barrel from a 357 max that would make a cool shooter, if it could be added to the mid frame.

While I cant comment on the cartridge fit on the mid frame I can say I had an original Vaquero (large frame) 45 colt barrel that fit straight up to a new vaquero, so Yes your barrel will fit.
